TheDetectX®humanST2EIAkitisdesignedtoquantitativelymeasureST2presentinavarietyofsamplesandtissueculturemedia.ArecombinanthumanST2standardisprovidedtogenerateastandardcurvefortheassay.StandardsordilutedsamplesarePipettedintoaclearmicrotiterplatecoatedwithamonoclonalantibodytocaptureST2. Aftera60minuteincubation,theplateiswashed.AbiotinylatedST2antibodyisaddedandtheplateincubatedforanadditional60minutes.Followingasecondwash,peroxidaseconjugatedstreptavidinisaddedandtheplateisincubatedfor30minutesandwashed.Substrateisthenaddedtotheplate,whichreactswiththeboundperoxidaseconjugatedstreptavidin.Afteranincubationtheintensityofthegeneratedcolorisreadat450nm.

ST2(alsoknownasgrowthSTimulationexpressedgene2,IL1RL1,DER4,T1andFIT‑1)isamemberoftheToll-like/IL-1-receptorsuperfamily. ClinicalandexperimentalobservationsledtotheassociationofST2withdiseasessuchasasthma,pulmonaryfibrosis,rheumatoidarthritis,collagenvasculardiseasesandsepticshock.SerumlevelsofST2areelevatedinpatientswithacutecasesofbronchialasthma,andinemergency‑roompatientspresentingwithshortnessofbreath.SerumlevelsofST2candiscriminatebetweenheartfailureandnon‑cardiovascularetiologies.
