TheDetectX^®Levonorgestrel(LNG)ImmunoassaykitisdesignedtoquantitativelymeasureLevonorgestrelpresentinserum,plasma,saliva,urine,driedfecalsamples,water,andtissueculturemediasamples.ThiskitmeasurestotalLNGinextractedserum,plasmaandfecalsamples.

Alevonorgestrelstocksolutionisprovidedtogenerateastandardcurvefortheassayandallsamplesshouldbereadoffthestandardcurve.StandardsordilutedsamplesarePipettedintoaclearmicrotiterplatecoatedwithanantibodytocapturerabbitantibodies.Alevonorgestrel-peroxidaseconjugateisaddedtothestandardsandsamplesinthewells.Thebindingreactionisinitiatedbytheadditionofarabbitpolyclonalantibodytolevonorgestreltoeachwell.Afteranhourincubationtheplateiswashedandsubstrateisadded.Thesubstratereactswiththeboundlevonorgestrel-peroxidaseconjugate.Afterashortincubation,thereactionisstoppedandtheintensityofthegeneratedcolorisdetectedinamicrotiterplatereadercapableofmeasuring450nmwavelength.Theconcentrationofthelevonorgestrelinthesampleiscalculated,aftermakingsuitablecorrectionforthedilutionofthesample,usingsoftwareavailablewithmostplatereaders.
