TheDetectX®GlutathioneS-Transferase(GST)FluorescentActivitykitisdesignedtoquantitativelymeasuretheactivityofGST.AGSTstandardisprovidedtogenerateastandardcurvefortheassay.Thekitutilizesanon-fluorescentmoleculethatisasubstratefortheGSTenzymewhichcovalentlyattachestoglutathione(GSH)toyieldahighlyfluorescentproduct.Intheendpointmode,mixingthesampleorstandardwiththesuppliedDetectionReagentandGSHyieldsafluorescentproductwhichisreadat460nminafluorescentplatereaderwithexcitationat390nm.Forratedetermination,theintensityat460nmisreadperiodicallyandplottedversustime.
TheDetectX®GlutathioneS-Transferase(GST)familyofisozymesfunctiontodetoxifyandneutralizeawidevarietyofelectrophilicmoleculesbymediatingtheirconjugationwithreducedglutathione.HumanGSTsareencodedby5genefamilies,expressinginalmostalltissuesasfourcytosolicandonemicrosomalforms.Dividingthefamilybyisoelectricpoints,thebasicalpha(pI8to11),theneutralmu(pI5to7)andacidicpi(pH<5)classesarepopulatedbyadditionalsubclasses,eachisozymedisplayingdifferentialspecificityforgivenelectrophilicmolecules.Givenitspivotalroleinamelioratingoxidativestress/damage,GSTactivityhasbeenrepeatedlyinvestigatedasabioMarkerforarthritis,asthma,COPD,andmultipleformsofcancer,aswellasanenvironmentalmarker.ExaminationofGSTisoformsandactivityinhumancancers,tumorsandtumorcelllineshasrevealedthepredominanceoftheacidicpiclass.FurThermore,thisactivityisthoughttosubstantiallycontributetotheinnateoracquiredresistanceofspecificneoplasmstoanticancertherapy.
