TheDetectX®GalactoseColorimetricDetectionKitisdesignedtoquantitativelymeasuregalactoseinavarietyofsamples. AD-(+)-galactosestandardisprovidedtogenerateastandardcurvefortheassayandallsamplesshouldbereadoffthestandardcurve.SamplesaremixedwiththeColorimetricSubstrateandhorserADIshperoxidaseandthereactioninitiatedbyadditionofgalactoseoxidase.Thereactionisincubatedatroomtemperaturefor30minutes.Thegalactoseoxidasereactswithgalactosetoproducehydrogenperoxidewhich,inthepresenceofHRP,reactswiththecolorlessColorimetricSubstratetoproduceapink-coloredproductwhichisreadat560nm.Increasinglevelsofgalactosecausealinearincreaseincolor.
Galactoseisahexosesugarthatdiffersfromglucoseonlybytheconfigurationofthehydroxylgroupatthecarbon-4position.Presentasananomericmixtureofα-D-galactoseandß-D-galactose,thismonosaccharideexistsabundantlyinmilk,dairyproductsandmanyotherfoodtypessuchasfruitsandvegetables.AbsorptionofgalactoseinhumansismediatedbytheNa+/glucoseco-transportersSGLT1andSGLT2fromfoodacrossthebrushbordermembraneoftheproximaljejunumandrenalepithelium.Othersourcesofgalactoseincludeendogenousproductionandnaturalturnoverofglycolipidsandglycoproteins.Adulthumanscanproduceupto2gramsofgalactoseperday.
