TheDetectX®Endothelin-1(ET-1)ImmunoassaykitisdesignedtoquantitativelymeasureET-1presentinavarietysamplesandtissueculturemedia.AnET-1standardisprovidedtogenerateastandardcurvefortheassayandallsamplesshouldbereadoffthestandardcurve.StandardsordilutedsamplesarePipettedintoaclearmicrotiterplatecoatedwithamonoclonalantibodytocaptureET-1presentinthesample.Aftera60minuteincubation,theplateiswashedandaperoxidaseconjugatedET-1antibodyisadded.Theplateisagainincubatedfor60minutesandwashed.Substrateisthenaddedtotheplate,whichreactswiththeboundET-1conjugatedantibody.Afterathirdincubation,thereactionisstoppedandtheintensityofthegeneratedcolorisdetectedinamicrotiterplatereadercapableofmeasuring450nmwavelength.TheconcentrationoftheET-1inthesampleisthencalculated.
