TheDetectX®Dehydroepiandrosteronesulfate(DHEA-S)ImmunoassaykitusesaspecificallygeneratedantibodytomeasureDHEA-Sinserum,plasma,urine,andsalivasamples. ThekitwillalsomeasureDHEA-Spresentinfecalextracts and tissueculturemediasamples. StandardsordilutedsamplesarePipettedintoaclearmicrotiterplatecoatedwithanantibodytocapturesheepantibodies.ADHEA-S-peroxidaseconjugateisaddedtothestandardsandsamplesinthewells.ThebindingreactionisinitiatedbytheadditionofapolyclonalantibodytoDHEA-Stoeachwell.Aftera2hourincubationtheplateiswashedandsubstrateisadded.ThesubstratereactswiththeboundDHEA-S-peroxidaseconjugate.Theintensityofthegeneratedcolorisdetectedinamicrotiterplatereaderat 450nm.

DHEA-Sisprimarilyproducedbytheadrenalglands and itisusefulasaMarkerforadrenalfunction.Adrenaltumors,cancers,andhyperplasiacanleadtotheoverproductionofDHEA-S.HighDHEA-Slevels canleadtoamenorrheaandvisIBLesymptomsofvirilization.Thesechangesvaryinseverityandmayinclude:adeepervoice,hirsutism,malepatternbaldness,muscularity,acneandenlargementoftheAdam’sapple.WomenwithpolycysticovarysyndrometendtohaveelevatedlevelsofDHEA-S.ExcesslevelsofDHEA-Sinchildrencancauseprecociouspubertyinboys;andambiguousexternalgenitalia,excessbodyhair,andabnormalmenstrualperiodsingirls.
