TheDetectX®CyclicAMP(cAMP)DirectImmunoassaykitisdesignedtoquantitativelymeasurecAMPpresentinlysedcells,tissue,plasma,urine,salivaandculturemediasamples.ThekitisuniqueinthatallsamplesandstandardsaredilutedintoanacidicSampleDiluentwhich isdesignedtolysecells,inactivatePDEenzymes,andstabilizecAMP. AcAMPstandardisprovidedtogenerateastandardcurvefortheassay.AspecialPlatePrimerisaddedtoallthewellsandstandardsorsamplesinSampleDiluentarePipettedintotheprimedclearmicrotiterplate.AcAMP-peroxidaseconjugateisaddedtothestandardsandsamplesinthewells.ThebindingreactionisinitiatedbytheadditionofanantibodytocAMP.After2hours,theplateiswashedandsubstrateisadded.ThesubstratereactswiththeboundcAMP-peroxidaseconjugate.Afterashortincubation,thereactionisstoppedandtheintensityofthegeneratedcolorisread.AnoptionalAcetylationFormatallowssuperlowconcentrationsofcAMPtobemeasured.
Adenosine-3’,5’-cyclicmonophosphate,orcyclicAMP(cAMP),isoneofthemostimportantsecondmessengersandakeyintracellularregulator.DiscoveredbySutherlandandRallin1957,itfunctionsasamediatorofactivityforanumberofhormones,includingepinephrine,glucagon,andACTH.AdenylatecyclaseisactivatedbythehormonesglucagonandadrenalineandbyGprotein.Liveradenylatecyclaserespondsmorestronglytoglucagon,andmuscleadenylatecyclaserespondsmorestronglytoadrenaline.cAMPdecompositionintoAMPiscatalyzedbytheenzymephosphodiesterase.IntheHumanMetabolomeDatabasethereare166metabolicenzymeslistedthatconvertcAMP.
