TheDetectX®Butyrylcholinesterase(BChE)FluorescentActivitykitisasimple,sensitiveandstablekitthatmeasuresBChEactivityinawidevarietyofsamples.Thekitcontainseverythingneededtomeasureactivity,includingthemicrotiterplate.Dilutedsamplesareaddedtothewells.ThereactionisinitiatedbyadditionofareactionmixcontainingthebutyrylthiocholinesubstrateandThioStar®.Aftera20minuteincubation,thesignalisreadat510nmwithexcitationat390nm.Theactivitycanalsobereadkinetically.AstablehumanBChEstandardsolutionisprovidedtogenerateastandardcurvefortheassay.TheassayshowsalmostzeroreactivitywithAChE.
Butyrylcholinesterase(BChE)belongstothesamestructuralclassofproteinsasacetylcholinesterase(AChE).The440kDatetramericglycoproteinispredominantlyfoundinblood,kidneys,intestine,liver,lung,heartandthecentralnervoussystem.Manyspecies,suchashuman,horseandmiceexhibithighBChEactivityinplasma,whereasratshavehigheracetylcholinesteraseactivityinplasma.AdeficiencyofBChEcanresultindelayedmetabolismofvariousdrugs,suchascocaine,andtreatmentwithdosesofBChEcanhelpinovercomingthephysiologicalreactiontothem.InAlzheimer’sdiseasetherapeutics,theuseofpotentcholinesteraseinhibitorsprotectthelimitedacetylcholinelevels,actingonbothAChEandBChE.BChEmayhaverolesinattention,executivefunction,emotionalmemoryandbehaviour.Otherintentionalorenvironmentalmethodsofimpairmentareorganophosphatesandcarbamateswithanticholinergicproperties,whichareusedasinsecticidesworldwideoraswarfareagents.
