AllopregnanoloneaidsneurogenesisandhasbeenfoundtoreverseneuronproliferativedeficitandcognitivedeficitsinamousemodelofAlzheimer’sdisease. AllopregnanolonehasalsobeenshowntorestorefunctionalityinamousemodelofParkinson’sdiseaseandtoimprovebehavioralproblemsinpost-traumaticstressdisorder.

TheDetectX^®AllopregnanoloneImmunoassaykitisdesignedtoquantitativelymeasureallopregnanolonepresentinextractedserum,plasma,driedfecalsamples,ordilutedurineandtissueculturemediasamples. Pleasereadthecompletekitinsertbeforeperformingthisassay. Anallopregnanolonestandardisprovidedtogenerateastandardcurvefortheassayandallsamplesshouldbereadoffthestandardcurve. StandardsordilutedsamplesarePipettedintoaclearmicrometerplatecoatedwithanantibodytocapturemouseantibodies. Anallopregnanolone-peroxidaseconjugateisaddedtothestandardsandsamplesinthewells. Thebindingreactionisinitiatedbytheadditionofamonoclonalantibodytoallopregnanolonetoeachwell.

Thekithastwoformatoptions:aprimaryincubationoftwohoursatroomtemperaturewithshaking,orovernightat4°C. Attheendoftheincubationperiodtheplateiswashedandsubstrateisadded. Thesubstratereactswiththeboundallopregnanolone-peroxidaseconjugate.Afterashortincubation,thereactionisstoppedandtheintensityofthegeneratedcolorisdetectedinamicrotiterplatereadercapableofmeasuring450nmwavelength. Theconcentrationoftheallopregnanoloneinthesampleiscalculated,aftermakingsuitablecorrectionforthedilutionofthesample,usingsoftwareavailablewithmostplatereaders.
