Anallopregnanolonestandardisprovidedtogenerateastandardcurvefortheassayandallsamplesshouldbereadoffthestandardcurve. StandardsorsamplesarePipettedintoawhitemicrotiterplatecoatedwithantibodytocapturerabbitantibodies. Anallopregnanolone-peroxidaseconjugateisaddedtothewellsandthebindingreactionisinitiatedbyadditionofapolyclonalantibodytoallopregnanolonetoeachwell. Theassayisincubatedovernightat4˚C. Attheendoftheincubationsperiodtheplateiswashedandchemiluminescentsubstrateisadded. Thechemiluminescentsubstratereactswiththeboundallopregnanolone-peroxidaseconjugatetogeneratelight.
