TheDetectX®Acetylcholinesterase(AChE)ActivitykitisdesignedtoquantitativelymeasureAChEactivityinavarietyofsamples.AhumanAChEstandardisprovidedtogenerateastandardcurvefortheassay.Thekitutilizesaproprietarynon-fluorescentmolecule,ThioStar®,thatcovalentlybindstothethiolproductofthereactionbetweentheAChESubstrateandAChEinthestandardsorsamples,yieldingafluorescentproductreadat510nminafluorescentplatereaderwithexcitationat390nm.
Acetylcholinesterases(AChE)appearcriticaltobothdevelopmentandfunctionofthenervoussystem.TheuseofAChEinhibitorsastherapeuticagentsandpesticideshasspurreddetailedinvestigationsofcholinesterasessincetheiridentificationbyDale.Acetylcholine(ACh)isanessentialneurotransmitterinthecentralandperipheralnervoussystems.Inthebrainmultipleareasexistwherecholinergicneuronsareconcentrated.NicotinicandmuscarinicAChreceptorsarerecognizedasbindingandeffectorproteinstomediatechemicalneurotransmissionatneurons,ganglia,heartandsmoothmusclefibersandglands.ThistrADItionalviewofAChEactingsolelyasneurotransmitterhastoberevisedbasedonthefindingspublishedbothearlyandlateinthelastcentury,demonstratingthenon-neuronalcholinergicsystem.
